According to Yamaha product literature and other data, both pianos use samples from a Yamaha CFIIIS, but the P140 uses 5 layer dynamic sampling for Grand Piano 1, while the P250 uses 3 layer dynamic sampling for Grand Piano 1. However, Yamaha might be counting the sustain and keyoff samples to get to a total of five for the P140 – thanks to SnazzyE for pointing this out. Additionally, the P250 has 128 note polyphony while the P140 has 64 note polyphony. The sound chips are also different. IMO, the P250 sounds much better. Hope this helps. Others may have additional information.

The P140 Owner's Manual (page 6). However, Yamaha might be counting the sustain and keyoff samples to get to a total of five. I modified my original post to include this.
